Does amlodipine increase cancer incidence?
نویسنده
چکیده
To the Editor: The Prospective Randomized Evaluation of the Vascular Effects of Norvasc Trial (PREVENT)1 amplifies the controversy over calcium antagonists and increased cancer risk.2 In this 36-month, placebo-controlled, randomized trial, the calcium antagonist amlodipine had no effect on the angiographic progression of coronary atherosclerosis or on the risk of major cardiovascular events. In the group assigned to amlodipine, the annualized rates of coronary artery bypass grafting and unstable angina were reduced by 1.0% and 2.1%, respectively. Unfortunately, incident cancers increased in the group assigned to amlodipine. The annualized rate of incident cancers was 1.8% in the amlodipine group and 1.0% in the placebo group. Thus, the 0.8% annualized increase in incident cancer in the amlodipine group was similar in magnitude but opposite in direction to the 1.0% annualized reduction in coronary artery bypass surgery. Because some observational data suggest that calcium antagonists may cause cancer,2 this finding in PREVENT warrants further investigation. From a public health perspective, for every 1 million patients taking amlodipine for 1 year, the incident cancer rate will increase by 8000 because of the drug. Perhaps we are trading one problem for another.

Comparative Analysis On Incidence Of Pedal Oedema Between Amlodipine, Cilnidipine And S-Amlodipine In Mild To Moderate Hypertensive Individuals Of Either Sex
Aim And Objective. According to JNC VIII guideline calcium channel blockers are first line of treatment in both general black or non black population (including those with diabetes) .It controls hypertension adequately, but very often produces ankle oedema. Our object of study was to compare the incidence of ankle oedema and its variation on the basis of gender , amongst commonly used Dihydropy...
متن کاملThe combination of amlodipine/valsartan 5/160 mg produces less peripheral oedema than amlodipine 10 mg in hypertensive patients not adequately controlled with amlodipine 5 mg
AIMS To demonstrate the benefit of the combination amlodipine/valsartan 5/160 mg over amlodipine 10 mg, in producing a lower incidence of peripheral oedema for a comparable mean sitting systolic blood pressure (MSSBP) reduction. METHODS After a 4-week amlodipine 5 mg run-in phase, inadequately controlled hypertension patients (aged > or = 55 years, MSSBP > or = 130 and < or = 160 mmHg) were r...
متن کاملRandomized study to compare valsartan ± HCTZ versus amlodipine ± HCTZ strategies to maximize blood pressure control
OBJECTIVE Delays in achieving blood pressure (BP) control may increase morbidity and mortality in patients with hypertension. Thus, deciding which antihypertensive agent to use and at what dosage, in addition to determining when to initiate combination therapy and which agents to combine, is important for achieving BP control. METHODS This randomized, double-blind, 14-week study was conducted...
متن کاملThe effect of amlodipine on respiratory and pulmonary vascular responses to hypoxia in mountaineers.
Calcium antagonists are known to reduce the incidence of high-altitude pulmonary oedema, but the mechanism is unclear. The aim of this study was to examine the effects of the calcium antagonist, amlodipine, on cardiac and respiratory responses in normoxia and hypoxia. Fourteen normal subjects aged 31+/-4 yrs who had climbed to altitudes of 5,000-7,500 m without problems were randomly assigned t...
متن کاملA current evaluation of the safety of angiotensin receptor blockers and direct renin inhibitors
The safety of angiotensin II receptor blockers (ARBs) for the treatment of hypertension and cardiovascular and renal diseases has been well documented in numerous randomized clinical trials involving thousands of patients. However, recent concerns have surfaced about possible links between ARBs and increased risks of myocardial infarction and cancer. Less is known about the safety of the direct...
